Microscopy Guide

You do not need an expensive lab to enjoy mushroom microscopy. Here is what you need and how to begin.

What you need

  • A compound microscope with 400x–1000x magnification (oil immersion helps at 1000x)
  • Blank glass slides and cover slips
  • A spore syringe
  • 70% isopropyl alcohol for wiping slides

Preparing a slide

  1. Shake the syringe gently to distribute the spores.
  2. Place one small drop on the center of a clean slide.
  3. Lower a cover slip onto the drop at an angle to avoid air bubbles.
  4. Start at low magnification to find the spores, then work up to 400x or 1000x.

What you will see

Spores vary by species — different shapes, sizes, colors, and surface textures. Comparing what you see against published descriptions is the heart of amateur taxonomy. Take notes and photos; over time you will start recognizing species on sight.
